In closely related plant species that display strong similarities in phenology and pollinator communities, differences in breeding system and associated shifts in floral traits may have important effects on the magnitude and direction of heterospecific pollen flow and hybridization. Here, we quantified the strength of several pre‐ and postzygotic barriers acting between the facultatively outcrossing Centaurium erythraea and the predominantly selfing C. littorale via a suite of experiments, and estimated the frequency of hybridization in the field using molecular markers. The reproductive barriers primarily responsible for preventing hybridization were essentially prezygotic and these acted asymmetrically. Due to differences in floral display, pollen production, and pollen transfer rates, heterospecific pollen flow occurred predominantly from C. erythraea to C. littorale. In C. littorale, on the other hand, close anther–stigma positioning and resulting higher capacity for autonomous selfing functioned as an efficient barrier to counterbalance the higher risk for hybrid mating. In both species the action of all reproductive barriers resulted in a small opportunity for hybrid establishment, which was confirmed by the occurrence of only ~1% putative hybrids in the field. The carcinogen 4-nitroquinoline 1-oxide (4NQO) and its metabolites undergo intercalative or covalent binding with DNA. Recent evidence indicates that the latter binding pattern is probably facilitated by an initial weaker intercalative interaction that can align potentially reactive sites on a 4NQO-metabolite and adjacent stacked bases. In the present study, we have proposed numerous possible covalent reaction products between 4NQO and its metabolites with DNA mini-helices based on chemical properties and key 'short-contacts' after energy-minimization in 21 different intercalative-like complexes. It is known from numerous experimental studies that 90% of the quinoline-bound DNAs in vivo involve guanine with the remaining 10% apparently involving adenine residues. The results of the present study suggest that this trend is not due to the greater affinity of the quinolines for guanine, but instead results from secondary processes involving the preferential formation of apurinic sites at aralkyl-adenine residues over that of aralkyl-guanine residues. In addition, observed mutational patterns can be rationalized in terms of the proposed reaction-products. The role of DNA repair mechanisms in the removal and correction of the different proposed reaction products are discussed. The binding pattern of several other aromatic carcinogens are similar to those depicted in the present work for the 4NQO-metabolites; hence the present study may be of some general significance. 相似文献

We describe here high-resolution spectra of etiolated leaves studied by means of selective monochromatic excitation of fluorescence as well as by hole-burning technique at liquid helium temperature. Up to now site selection spectroscopy has not been successfully applied to biologically active chromophores in vivo. Our results demonstrate that even in the presence of specific pigment-protein and pigment-pigment interactions very narrow purely electronic lines can be obtained in the optical spectra of biological systems. 相似文献

Stone tool‐use and manufacture is seen as an important skill during the evolution of our species and recently there has been increased interest in the understanding of perceptual‐motor abilities underlying this skill. This study provides further information with respect to the motor strategies used during stone knapping. Kinematics of the striking arm were recorded in expert and novice knappers while producing flakes of two different sizes. Using Uncontrolled Manifold Analysis, the results showed that knappers structure joint angle movements such that the hammer trajectory variability is minimized across trials, with experts displaying significantly smaller variability compared with novices. Principal component analysis further revealed that a single component captures the complexity of the strike and that the strike is governed by movements of the elbow and the wrist. Analysis of movement velocities indicated that both groups adjusted movement velocities according to flake size although experts used smaller hammer, wrist, and elbow velocities in both flake conditions compared with novices. The results suggest that while the gross striking movement is easy to replicate for a novice knapper, it requires prolonged training before a knapper becomes attuned to the finer details necessary for controlled flaking. © 2013 Wiley Periodicals, Inc. 相似文献
